摘要

目的 建立以反相高效液相色谱-蒸发光散射检测法(RP-HPLC-ELSD)测定不同产地蕤仁中亚油酸含量的测定方法,为有效控制蕤仁饮片的质量提供科学依据。 方法 采用回流法提取亚油酸;色谱条件为: AichromaBond-AQ C18色谱柱(4.6 mm× 250 mm, 5 μm);流动相:乙腈-0.1%甲酸水 (90∶10);流速:1.0 mL·min-1;柱温:25 ℃;ELSD检测器。 结果 亚油酸对照品在0.998 9~49.945 μg内线性关系良好 (r=0.998 4),平均回收率为 100.1%,RSD 为 1.2%。结论 本方法准确,重复性好,简便易行,适用于蕤仁中亚油酸的含量测定。

Abstract

OBJECTIVE To develop a RP-HPLC-ELSD method for determining linolic acid in Prinsepiae Nux from different habitats and provide the basis for controlling the herb quality. METHODS Reflux extraction method was used to isolate linolic acid from Prinsepiae Nux and RP-HPLC-ELSD method was used to determine the content of linolic acid. RESULTS The calibration curves of linolic acid had good linearity over the range of 0.998 9-49.945 μg (r=0.998 4), and the average recovery of linolic acid was 100.1% with RSD of 1.2%. CONCLUSION The method is simple, sensitive and accurate, which is suitable for the determination of linolic acid in Prinsepiae Nux.
